Description

MRC ARL award, This research will fill the existing knowledge gap on how the brain is injured in cryptococcal meningitis. This models and methods would then serve as a platform for studying the mechanisms of other infections of the Central Nervous System that are caused by bacteria, parasites and viruses to inform the development of the much-needed new therapies.

Objectives

AIM 1: Determine the neuroimmune response of microglia and neurons to C. neoformans Objective 1.1: Characterise neuroimmune signalling pathways in microglial cells during C. neoformans infection Objective 1.2: Characterise the mechanisms and effects of neuronal invasion by C. neoformans Objective 1.3: Characterise cell-type specific gene expression during C. neoformans infection in organotypic slices using single-nucleus RNA sequencing AIM 2: Investigate the effect of cryptococcal entry and accumulation in brain compartments on fluid flow and neuroimmune homeostasis Objective 2.1: A spatiotemporal anatomopathological characterisation of cryptococcal infection of the mouse and human brain Objective 2.2: An investigation of the effect of cryptococcal infection on fluid transport in a mouse model of CM Objective 2.3: Characterise spatial gene expression profiles during C. neoformans infection in an in vivo rodent model of CM using spatial transcriptomic sequencing
